METHODS: Nineteen canines underwent bilateral transection all coccygeal sacral (S) spinal roots, dorsal roots lumbar (L)7 hypogastric nerves. After exclusions, 8 were reinnervated 12 months post-decentralization with obturator-to-pelvic sciatic-to-pudendal nerve transfers then euthanized 8-12 later; four served as decentralized only animals. Controls included six sham-operated three unoperated Bladder tissues assessed for contractile responses potassium chloride (KCl) electric field stimulation (EFS) before after receptor desensitization alpha, beta-methylene adenosine triphosphate (α,β-mATP), antagonism atropine, or sodium channel blockade tetrodotoxin. RESULTS: Atropine inhibition EFS-induced contractions increased 26%in 34%in animals compared controls. Maximal α,β-mATP did not differ between groups. In strips from animals, response EFS was enhanced 52% at lower frequencies KCl normal CONCLUSIONS: The observation nerve-evoked antagonist no change in responsiveness agonist suggests either decreased ATP release ectoATPase activity detrusor a consequence decentralization. reduction frequency required produce maximum contraction following may be due sensitivity effectiveness neurotransmission. Transient contractions of urinary bladder smooth muscle are drivers of afferent nerve activity during filling
Activation of afferent nerves during urinary bladder (UB) filling conveys the sensation of UB fullness to the central nervous system (CNS). Although this sensory outflow is presumed to reflect graded increases in pressure associated with filling, UBs also exhibit nonvoiding, transient contractions (TCs) that cause small, rapid increases in intravesical pressure. Here, using an ex vivo mouse bla...
متن کاملNegative feedback regulation of nerve-mediated contractions by KCa channels in mouse urinary bladder smooth muscle.
When the urinary bladder is full, activation of parasympathetic nerves causes release of neurotransmitters that induce forceful contraction of the detrusor muscle, leading to urine voiding. The roles of ion channels that regulate contractility of urinary bladder smooth muscle (UBSM) in response to activation of parasympathetic nerves are not well known. The present study was designed to charact...
متن کاملNeurogenic Bladder Part 2 of Bladder Dysfunction
Neurogenic bladder , explanations follow in the text . i. Urge Incontinence manifesting as urgency and the urinary leakage even before individual reaches the toilet. ii. Over flow incontinence manifesting as involuntary release of urine from an overfull urinary bladder, often in the absence of any urge to urinate. iii. Dribbling incontinence , the patient is never dry a continuous leakage of ur...
متن کاملINHIBITORY EFFECT OF KETOTIFEN ON CONTRACTIONS OF RAT ISOLATED BLADDER
Ketotifen is a benzocycloheptathiophene with a range of pharmacological activities. The present study was carried out to evaluate the action of ketotifen on isolated rat bladder contractions induced by KCI and acetylcholine, compared with the effects of other drugs. Ketotifen (5 µM) reduced the response to acetylcholine on rat isolated bladder without altering the maximum response and shift...
متن کاملAB295. SPR-22 Functional relevance of purinergic P2X4R in bladder smooth muscle
Objective: Under physiologic conditions in animals and pathologic conditions in humans, purinergic mechanisms contribute significantly to detrusor contractions. Although the activation of P2X1 receptors (P2X1R) accounts for the largest portion of bladder smooth muscle (BSM) responses to ATP, previous reports have shown that P2X1R antagonists do not completely abolish the purinergic component of...
